Plasma biomarkers associated with adverse outcomes in patients with calcific aortic stenosis

Vidula MK, Orlenko A, Zhao L, et al. - Findings demonstrate strong associations of plasma biomarkers with the risk of adverse outcomes in patients suffering from aortic stenosis (AS). Most strong links with prognosis were shown by biomarkers of inflammation and calcification.

  • This study involved 708 patients with calcific AS in whom 49 biomarkers were measured using a Luminex platform.

  • Post-adjustment for confounders, death was significantly predicted by multiple biomarkers including TNF-α, TNFRI (TNFRSF1A), fibroblast growth factor (FGF)-23, NT-proBNP, MMP-7, syndecan-1, ST2, IL-8, pentraxin (PTX)-3, NGAL, osteoprotegerin (OPG), and endostatin.

  • Adjusted analyses showed various biomarkers could significantly predict DHFA, including FGF-23, TNF-α, TNFRI, angiopoietin-2, syndecan-1, ST2, IL-8, PTX-3, OPG, and NT-proBNP.

  • Strong links of machine-learning-multimarker models with adverse outcomes were evident, and IL-6 and FGF-23 were the biomarkers of highest importance in these models.
