Melanomas with activating RAF1 fusions: Clinical, histopathologic, and molecular profiles

Williams CEA, Shah N, Montesion M, et al. - Utilizing the case archive of clinical samples with comprehensive genomic profiling (CGP) by a hybrid capture-based DNA sequencing platform, researchers tried to find out this distinctive group of melanomas, they searched for melanomas with activating structural variants in RAF1 . For each case, clinical data, pathology reports, and histopathology were examined. They defined RAF1 breakpoints, fusion partners, and co-occurring genetic alterations. They found 40 cases (0.6%) featured fusions that created activating structural variants in RAF1 from a cohort of 7119 melanomas. The study found that activating RAF1 fusions define a significant subset of triple wild-type melanoma (2.1%) with frequent accompanying mutations in TERTp and CDKN2A . It was noted that CGP of melanomas may enhance tumor classification and inform potential therapeutic options, such as evidence of specific kinase inhibitors.
