Genetic variant associated with survival of patients with stage II-III colon cancer

Penney KL, Banbury BL, Bien S, et al. - Since many genetic variants were correlated with colorectal cancer risk, although few were related to survival times of patients, researchers conducted a genome-wide association study to detect variants linked to colon cancer survival time. They conducted a post-hoc analysis of data from NCCTG N0147 (Alliance), a randomized phase 3 trial of patients with resected stage III colon cancer, and from NSABP C-08 (NRG), a phase 3 trial that contrasted therapy regimens for patients with resected stage II or III colon cancer. A locus on chromosome 7p15.2 was significantly linked to overall survival time. The most important variant at this locus, rs76766811, is common among African Americans but uncommon among European Americans. This variant was related to times of overall survival and disease-free survival in only African Americans within strata of self-reported ancestry. For additional study populations, this result should be tested.
