HMGA2 and MED12 alterations frequently co-occur in uterine leiomyomas

Galindo LJ, et al. - Experts searched for the mutation in MED12 and evaluated the expression of HMGA2 in 20 uterine leiomyomas and their matched myometrium. The former studies, contrary to the present findings, demonstrated that mutations in MED12 and overexpression of HMGA2 are mutually exclusive. Here, it was seen that a common phenomenon in fibroids was overexpression of HMGA2 mRNA in tumors measured by quantitative PCR and compared to myometrium and it was seen to be frequently associated with MED12 mutations. Furthermore, in leiomyoma tumorigenesis up-regulation of HMGA2 as an early event was indicated by common clonal origin of tumors overexpressing HMGA2 mRNA and its expression in few myometrial tissue.
