Distinct polygenic score profiles in schizophrenia subgroups with different trajectories of cognitive development

Dickinson D, Zaidman SR, Giangrande EJ, et al. - Researchers analyzed a schizophrenia sample in order to derive as well as to characterize cognitive development trajectory subgroups, and they also profiled the subgroups across polygenic scores (PGSs) for schizophrenia, cognition, educational attainment, and attention deficit hyperactivity disorder. They analyzed data from 540 schizophrenia patients, 247 unaffected siblings, and 844 controls, from the National Institute of Mental Health. In the schizophrenia group, three cognitive trajectory subgroups were found via cluster analyses: preadolescent cognitive impairment (19%), adolescent disruption of cognitive development (44%), and cognitively stable adolescent development (37%). Experts noted that 7.9% of the variance in subgroup membership was significantly predicted by the four PGSs, collectively. The best adult clinical results were evident in cognitively stable people and these individuals, when compared with controls, demonstrated a difference only in schizophrenia PGSs. The most severe symptoms post-diagnosis were seen in those with adolescent disruption of cognitive development, and these individuals showed cognitive impairment. As per the findings, different premorbid as well as clinical features were demonstrated by subgroups derived from patterns of premorbid and current IQ, and these features converged with wide genetic profiles. A likely contribution of the simultaneous study of multiple PGSs to valuable clinical stratification in schizophrenia was also suggested.
