Cytokine-like 1 is a novel proangiogenic factor secreted by and mediating functions of endothelial progenitor cells

Schneller D, et al. - Researchers evaluated the functions of cytokine-like 1 (CYTL1), a factor preferentially produced by endothelial colony forming cells (ECFCs), in terms of vessel formation. They observed that the factor not only increased angiogenic sprouting capabilities of ECFCs but is also capable of stimulating sprouting of mature endothelial cells, as demonstrated by modulation of expression. Also, it was reported that hypoxia could trigger CYTL1 and that CYTL1 functions largely independently of VEGF-A (vascular endothelial growth factor-A). CYTL1 was specifically O-glycosylated on two neighboring threonines in the C-terminal part, as confirmed on mass spectroscopy, and this modification was vital for its proangiogenic bioactivity. Upregulation of proinflammatory genes was not a function of CYTL1, but it strongly induced several metallothionein genes encoding anti-inflammatory and antiapoptotic proteins. Overall, proangiogenic functions ascribed to endothelial progenitors such as ECFCs could be mediated by CYTL1 in vivo. CYTL1 showed potential as a candidate to support vessel formation and tissue regeneration in ischemic pathologies.
