Cancer risk in families With Amsterdam criteria for Lynch syndrome

Sammader NJ et al. – A number of the cancers known to be associated with Lynch syndrome, several unreported cancer associations, and the risk of colorectal and endometrial cancers were markedly increased in first-, second-, and third-degree relatives. 

Methods

  • 202 families with Amsterday I and II criteria-positive pedigrees were studied.

Results

  • The following cancers were shown to be in excess in first-degree relatives with Amsterdam criteria pediagrees: colorectal (standardized morbidity ratio [SMR], 10.10); endometrial (SMR, 5.89); stomach (SMR, 2.90); small intestine (SMR, 7.72); prostate (SMR, 1.94); kidney (SMR, 3.22); urinary bladder (SMR, 1.62); thyroid (SMR, 2.26); and non-Hodgkin lymphoma (SMR, 2.10).
  • The risk of colorectal and endometrial cancers was increased in second-degree relatives (SMR = 4.31 and 2.70, respectively) and first cousins (SMR = 1.85 and 1.50, respectively).
