Biomarker immunoprofile and molecular characteristics in salivary duct carcinoma: Clinicopathologic and prognostic implications

Santana T, Pavel A, Martinek P, et al. - Clinicopathological and follow-up information of 25 cases diagnosed as salivary duct carcinoma (SDC) were gathered by the researchers in order to examine the prognostic implications of biomarker immunoprofile in a cohort of SDC and to recognize molecular features through next-generation sequencing (NGS) in a subset of cases. AR, GATA3, and CK5/6 were expressed by most cases and 42% expressed HER-2/neu. In 7/15 cases, ERBB2 gene amplification was proven by FISH. Apocrine HER2 (AR+/HER2+) subtype was significantly correlated with lower OS. In seven SDC cases, NGS analysis showed nine pathogenic mutations, and the most commonly mutated gene was HRAS, followed by PIK3CA and TP53. Homozygous deletion of locus 9p21 (CDKN2A) and MDM2 amplification were exhibited by one case, for each, respectively. Hence, in SDC, Apocrine HER-2 (AR+/HER2+) was concluded as a potential biomarker of poor outcome. Moreover, NGS analysis showed recurrent mutations in SDC.
