Effect of folate supplementation on insulin sensitivity and type 2 diabetes: A meta-analysis of randomized controlled trials

Lind MV, et al. - Researchers determined the impact of folate supplementation on insulin resistance (IR) and type 2 diabetes (T2D) outcomes by analyzing 29 relevant randomized controlled trials (RCTs; n=22,250), which were identified via a search of PubMed, Web of Science, and EMBASE databases, as well as prior systematic reviews and meta-analyses. These studies assessed fasting glucose, insulin, homeostasis model assessment for insulin resistance (HOMA-IR), glycated hemoglobin (HbA1c), or risk of T2D in relation to placebo-controlled folate supplementation alone or in combination with other B vitamins. Lowered fasting insulin and HOMA-IR were noted to be a result of folate supplementation vs placebo, but no overall impact was observed for fasting glucose or HbA1c. Findings revealed correlation between changes in homocysteine after folate supplementation and changes in fasting glucose and HbA1c. Risk of T2D in relation to folate supplementation was assessed in only two studies, and no change in RR was observed. For glucose homeostasis and lowering IR, folate supplementation might be a beneficial strategy. Due to insufficient data, no conclusions could be drawn regarding the impact on T2D development.
