Circulating levels of Dickkopf-related protein 1 decrease as measured GFR declines and are associated with PTH levels

Forster CM, White CA, Turner ME, et al. - Given that in the development of adynamic bone disease in early-stage chronic kidney disease, the Wnt/β-catenin pathway has been implicated, and that dickkopf-related protein 1 (DKK1) and sclerostin represent antagonists of the Wnt/β-catenin pathway, researchers herein sought to characterize concentrations of DKK1, sclerostin, and other biomarkers of mineral metabolism among participants (n = 90) across a spectrum of inulin-measured glomerular filtration rate (GFR). Findings revealed opposing trends in DKK1 and sclerostin (Wnt/β-catenin pathway inhibitors) as mGFR (GFR measured by urinary inulin clearance) reduced. Unlike sclerostin, DKK1 concentrations reduced significantly as mGFR decreased and was found to be independently related to parathyroid hormone.
