Neuropilin-2 is an independent prognostic factor for shorter cancer-specific survival in patients with acinar adenocarcinoma of the prostate

Borkowetz A, Froehner M, Rauner M, et al. - A tissue microarray with prostate tissue specimens from 400 individuals with prostate cancer (PCa) treated by radical prostatectomy was generated in order to assess the staining of neuropilin-2 (NRP2) and vascular endothelial growth factor C (VEGFC) in primary PCa tissue, to examine the role of NRP2 as a possible predictor of unfavorable PCa outcome and to investigate the role of v-ets avian erythroblastosis virus E26 oncogene homolog (ERG) alone and in combination with NRP2 staining as prognostic marker in this PCa cohort. For cancer-specific survival (CSS), NRP2 appeared as an independent prognostic factor. Furthermore, in individuals at high risk for progression, the relationship between NRP2 expression and shorter CSS was also particularly proclaimed. The relationship between NRP2 and the transmembrane protease serine 2-ERG gene fusion status evaluated by immunohistochemical nuclear ERG staining was assessed. Nonetheless, ERG staining alone did not reveal any prognostic importance. In ERG-negative tumors, NRP2 immunostaining is significantly correlated with shorter CSS. In ERG-positive tumors, no prognostic influence of NRP2 expression on CSS was noted. Hence, this study for a worse clinical outcome recognizes NRP2 as a significant prognostic marker particularly in individuals with a high-risk PCa and in individuals with ERG-negative PCa.
