Penicillin V four times daily for five days vs three times daily for 10 days in patients with pharyngotonsillitis caused by group A streptococci: Randomized controlled, open label, non-inferiority study

Ståhlgren GS, et al. - Via performing an open-label, randomized, controlled, non-inferiority study in 17 primary healthcare centers in Sweden between September 2015 and February 2018, researchers ascertained whether total exposure to penicillin V could be lowered while maintaining sufficient clinical efficiency when treating pharyngotonsillitis caused by group A Streptococci. A total of 433 people were randomized to a 5-day regimen (n = 215) or 10-day regimen (n = 218). In individuals with pharyngotonsillitis caused by group A Streptococci, in terms of clinical outcome, penicillin V four times daily for 5 days was non-inferior to penicillin V three times daily for 10 days. Between the two intervention groups, no variation in the number of relapses and complications was noted. Moreover, 5-day treatment with penicillin V four times daily could be an option relative to the currently supported 10-day regimen.
