Mendelian randomization study for genetically predicted polyunsaturated fatty acids levels on overall cancer risk and mortality

Liyanage UE, et al. - Using Mendelian randomization (MR), researchers analyzed genetic evidence to ascertain a causal role for polyunsaturated fatty acids (PUFAs) on overall cancer outcomes. Using previous literature, they recognized genetic instruments for six PUFAs and assessed their link with overall cancer risk (46,155 cases, 270,342 controls) and cancer mortality (6,998 deaths, 270,342 controls) in the UK Biobank cohort. To combine SNP-estimates, they used the inverse variance weighted model. They derived log estimates per SD change in each PUFA. The hypothesis that PUFAs reduce overall cancer risk or mortality was not supported in this study. An increased risk for colorectal cancer was reported in relation to higher AA levels.
